A multigenic construct that expresses a a truncated Hsap\MYB protein and a hairpin cluster, each downstream of their own inducible UAS promoter. The truncated Hsap\MYB protein was found in a patient with adenoid cystic carcinoma - it lacks the C-terminal cytoplasmic domain required to regulate Hsap\MYB activity. The hairpin cluster (version 12.2) targets orthologs of human ERCC2, FAT4, and FAT1/3 (Xpd, ft and kug, respectively), which harbored misssense mutations in the patient's tumor.
The expression of Hsap\MYBΔC.UAS, XpddsRNA.UAS.012.2, ftdsRNA.UAS.012.2 and kugdsRNA.UAS.012.2 under the control of Scer\GAL4ptc-559.1 induces an expansion of the ptc-positive domain of the wing disc's anterior/posterior boundary and a loss of the sharp boundary.
