Expression of Abd-BUAS.cCa under the control of Scer\GAL4insc-Mz1407 from mid third instar stage, in combination with a Gal80[ts] transgene, results in a reduced number of neuroblasts in the thoracic segments of the late third instar (male or female) larval ventral nerve cord, compared to controls.

The adulthood-only expression of Abd-BUAS.cCa (using Gal80[ts]) driven by Scer\GAL4C587, but not by Scer\GAL4VP16.nos.UTR or Scer\GAL4hh-Gal4, leads to adult testes exhibiting a significant increase in the number of cyst stem cells (and their localization away from the hub), a reciprocal decrease in the number of differentiated cyst cells, an increase in the number of early stage germline cells, and a strong increase in the number of DNA replicating cells at the apical testis tip (even away from the hub), as compared to controls. This Scer\GAL4C587-dependent expression also leads to a cell non-autonomous induction of both somatic (Zfh1) and germline (Vasa) lineage markers in some somatic-derived cells, as compared to controls.

Expression of Abd-BScer\UAS.cCa under the control of Scer\GAL4rn.PU suppresses wing formation.

Expression of Abd-BScer\UAS.cCa under the control of Scer\GAL469B in the trunk segments results in the formation of ectopic spiracles.

When Abd-BScer\UAS.cCa is driven by Scer\GAL4sd-SG29.1 abnormal wing vein patterns and incisions in the wing margin are seen.

Expression of Abd-BScer\UAS.cCa under the control of Scer\GAL4dpp.blk1 results in transformation of the arista to tarsus.

Embryos expressing Abd-BScer\UAS.cCa under the control of Scer\GAL4twi.PB form ectopic clusters of male-specific somatic gonadal precursor (msSGP) cells in more anterior regions of the embryo than normal. Gonads do not form in these embryos and germ cells are either scattered or associated with msSGPs.

79% of anterior dMP2 neurons survive in late stage embryos expressing Abd-BScer\UAS.cCa under the control of Scer\GAL4Vap.P0201 (in contrast to wild-type embryos, where these neurons are lost by the late embryonic stage). 100% of anterior dMP2 neurons survive in late stage embryos expressing Abd-BScer\UAS.cCa under the control of Scer\GAL4elav-C155. Expression of Abd-BScer\UAS.cCa under the control of Scer\GAL4elav-C155 in late stage Abd-BM2/Abd-BM5 embryos results in the suppression of cell death normally seen in anterior dMP2 and MP1 neurons in wild-type late stage embryos.

Expression of Abd-BScer\UAS.cCa under the control of Scer\GAL4sca-537.4 results in a mutant phenotype in the embryonic tritocerebrum. The phenotype has a penetrance of more than 95%.

Expression of Abd-BScer\UAS.cCa in the mesoderm under the control of Scer\GAL4unspecified results in severe defects in muscle development; the skeletal muscle pattern is massively deranged and there appear to be no dorsal vessel cells (based upon staining for Mhc expression).

Expression of Abd-BScer\UAS.cCa under the control of Scer\GAL4lab.PH does not result in morphological defects in the tritocerebrum or any other part of the embryonic brain.

Expression of Abd-BScer\UAS.cCa (driven by Scer\GAL4dpp.blk1) results in inhibition of eye development by inducing apoptosis.

Scer\GAL469B-mediated expression in wild type causes a pair of posterior spiracles to develop in all trunk segments.

Scer\GAL4E132 induced expression in the eye-antennal disc causes transformation of the antenna into leg. Scer\GAL4Bx-MS1096 or Scer\GAL4C-765 induced expression in the wing disc causes transformation of the wing into haltere-like tissue. Scer\GAL4C-765 induced expression also causes malformed notum, fused scutellum and disorganised pattern of bristles. Scer\GAL4c739 and Scer\GAL4C-765 induced expression in the abdomen transforms A1 into a posterior abdominal segment.

Scer\GAL4twi.PG induced expression blocks the formation of the gonad primordium.

When driven by Scer\GAL4Kr.PM, Scer\GAL4h-1J3 or Scer\GAL469B, in a wild type background, Abd-BScer\UAS.cCa generates ectopic primordia for the posterior spiracle. These go on to develop filzkorper and spiracular hairs. Keilin's organs are suppressed in the Abd-BScer\UAS.cCa, Scer\GAL4h-1J3 or Abd-BScer\UAS.cCa, Scer\GAL4Kr.PM combination.

Addition of Abd-BScer\UAS.cCa to Dfd16;Scr4 double mutant embryos (under the control of Scer\GAL4salm-459.2) results in both the Mx and Lb segments forming tracheal tubes instead of migratory gland primordia.

Co-expression of pbScer\UAS.T:Avic\GFP-EGFP suppresses the arista-to-tarsus transformation seen in flies expressing Abd-BScer\UAS.cCa under the control of Scer\GAL4dpp.blk1.

Embryos coexpressing abd-AScer\UAS.cGa and Abd-BScer\UAS.cCa under the control of Scer\GAL4twi.PB have ectopic male-specific somatic gonadal precursor (msSGP) cells and ectopic somatic gonadal precursor (SGP) cells. A large group of SGPs and msSGPs associates with most of the germ cells in the region where the gonad would normally be coalescing and ectopic SGPs and msSGPs also associate with one another in more anterior regions.

Expression of Abd-BScer\UAS.cCa under the control of Scer\GAL4lab.PH does not rescue the tritocerebral defects seen in lab14 embryonic brains.

Scer\GAL469B-mediated expression of Abd-BScer\UAS.cCa in linG1 mutants does not cause ectopic posterior spiracles development in the trunk segments.

Ubxabx-1 Ubxbx-3 Ubxpbx-1/Ubxlac1 combination transforms the metathorax into mesothorax (including transformation of halteres into wings), Scer\GAL4Bx-MS1096 induced expression allows partial rescue.

Scer\GAL4twi.PG induced expression of Abd-BScer\UAS.cCa blocks formation of the gonadal primordium, coexpression of Abd-BScer\UAS.cCa and abd-AScer\UAS.cGa restores gonadal mesoderm.

Comments

The loss of posterior dMP2 neurons that is seen in late stage Abd-BM2/Abd-BM5 embryos is rescued by expression of Abd-BScer\UAS.cCa under the control of Scer\GAL4Vap.P0201.
