Expressing exUAS.cBa under the control of Scer\GAL4GMR.PU induces ectopic cell apoptosis (in TUNEL assay) in the wing disc posterior compartment.

Ectopic expression of exScer\UAS.cBa under the control of Scer\GAL4nub.PU results in severely undergrown wings and expression under Scer\GAL4GMR.PU results in loss of eye tissue and smaller eyes in adult flies.

Expression of exScer\UAS.cBa under the control of Scer\GAL4C587 has no apparent germarium phenotype.

Expression of exScer\UAS.cBa under the control of Scer\GAL4GMR.PF induces a stripe of dying cells posterior to the morphogenetic furrow in third instar eye discs.

Expression of exScer\UAS.cBa under the control of Scer\GAL432B results in a substantial reduction in wing size.

Cells in the wing disc expressing exScer\UAS.cBa under the control of Scer\GAL4hh-Gal4 grow very poorly.

Expression of exScer\UAS.cBa under the control of Scer\GAL4GMR.PF leads to results in severe defects in eye development - adult eyes are blistered over the entire surface and are reduced in size. Expression of exScer\UAS.cBa under the control of Scer\GAL4hs.2sev results in severe defects in eye development - adult eyes are blistered and reduced in size. Blistering is visible in anterior regions but morphology is relatively normal in posterior ommatidial rows. The blistered areas show highly disorganised ommatidia. The non-blistered areas exhibit a lack of pigment cells, defects in photoreceptor recruitment and defects in planar polarity. Expression of exScer\UAS.cBa under the control of Scer\GAL4ey.PH produces defects in the eye disc. Discs are not flat and small relative to the antennal portions of the same disc. However, ommatidial preclusters look fairly well ordered. Expression of exScer\UAS.cBa under the control of Scer\GAL432B in eye discs leads to normal alignment of photoreceptor precursor pairs, but misrotated clusters are observed even in more anterior rows. Rotation defects are also prominent in anterior rows if exScer\UAS.cBa is driven by Scer\GAL4hs.2sev or Scer\GAL4GMR.PF. In Scer\GAL4hs.2sev, exScer\UAS.cBa discs one of the R3/R4 precursor pair is occasionally missing. The number of accessory cells which normally surround the photoreceptors appears to be greatly reduced. Organisation of ommatidia is severely disrupted and in many cases ommatidial preclusters have too few cone cell nuclei. Expression of exScer\UAS.cBa under the control of Scer\GAL4hs.2sev, Scer\GAL4GMR.PF or Scer\GAL4ey.PH causes an increase in apoptosis (as indicated by incorporation of Acridine Orange) in the eye disc. Expression of exScer\UAS.cBa under the control of Scer\GAL4bi-omb-Gal4 results in increased cell death in the wing pouch and parts of the leg disc. producing adult flies with severely reduced wings with patterning defects including loss of vein material and disruption of the margin. Legs appear to be fragile and twisted. Expression of exScer\UAS.cBa under the control of Scer\GAL4Bx-MS1096 results in small, unpatterned wings and misshapen legs.

When expression is driven by Scer\GAL432B in an otherwise wild type background the wing area is severely reduced. There is greater size reduction between L2 and L3 than anywhere else. Reduction in size is due to reduced cell number, not size. Some wings show ectopic anterior crossveins, with delta-like wing veins and notched wing margins appearing at lower frequencies.

Heterozygosity for wtsx1 partially suppresses the reduction in size and patterning defects seen in the eyes of exScer\UAS.cBa; Scer\GAL4GMR.PF animals.

The increase in cell death the late third instar eye discs of exScer\UAS.cBa; Scer\GAL4GMR.PF animals is largely suppressed by wtsx1/wtsx1 or hpo42-47/hpo42-47.

Overexpression of exScer\UAS.cBa in the developing eyes of ft422 mutant heads, under the control of Scer\GAL4GMR.PF results in small and rough eyes, very similar to the overexpression of exScer\UAS.cBa in a wild-type background, indicating that exScer\UAS.cBa expression suppresses the ft422 mutant eye phenotype. ft422 mutant phenotypes, such as extra vibrassae, are still observed in the head cuticle, where exScer\UAS.cBa is not overexpressed (under the control of Scer\GAL4GMR.PF).

Co-expression of banEP3622 almost completely suppresses the eye phenotype caused by expression of exScer\UAS.cBa under the control of Scer\GAL4hs.2sev. The eye phenotype caused by expression of exScer\UAS.cBa under the control of Scer\GAL4hs.2sev is enhanced by Df(3L)banΔ1/+. Co-expression of both CycDScer\UAS.cMa and Cdk4Scer\UAS.cMa in animals expressing exScer\UAS.cBa under the control of Scer\GAL4hs.2sev increases the overall size of the eye but has little effect on the roughness and blistering. The eye phenotype caused by expression of exScer\UAS.cBa under the control of Scer\GAL4hs.2sev is not altered by Cdk43/+.

Addition of BacA\p35Scer\UAS.cHa to exScer\UAS.cBa, Scer\GAL4hs.2sev flies rescues the phenotypes seen in exScer\UAS.cBa, Scer\GAL4hs.2sev flies alone. Externally the eye is much smoother and in sections most of the cell loss is rescued, although occasional photoreceptor and pigment cell loss is evident. Defects in planar polarity are still prominent.
