Expression of Tak1Scer\UAS.cTa under the control of Scer\GAL4wor.PA induces neuroblast loss in larval brains.

Overexpression of Tak1Scer\UAS.cTa under the control of Scer\GAL4GMR.PF, induces autophagy in developing eye discs. This results in a lack of eye pigmentation and a rough, impaired appearance compared with control flies.

Scer\GAL4GMR.PF-driven expression of Tak1Scer\UAS.cTa results in a non-pigmented and small eye and lethality of 45% of differentiated pupae.

Expression of Tak1Scer\UAS.cTa under the control of Scer\GAL4GMR.PF results in a small eye phenotype.

When expression is driven by Scer\GAL4prd.RG1, anterior rows of denticles are lost in even numbered segments.

most Tak1Scer\UAS.cTa,Scer\GAL4GMR.PF animals die at pupal stages when grown at 25oC. They exhibit a small eye phenotype. The number of apoptotic cells increases dramatically in regions posterior to the morphogenetic furrow in the developing eye. Sequential recruitment of the photoreceptor cells is disrupted in mutant eye discs. Early photoreceptor induction appears to occur normally but later photoreceptor induction is delayed or disrupted and photoreceptor clustering shows an abnormal and diffuse pattern.

The number of autolysosomes, indicating the level of autophagy, is increased upon co-expression of S6kKQ.Scer\UAS and Tak1Scer\UAS.cTa in the developing eye disc, under the control of Scer\GAL4GMR.PF, compared to expression of either transgene alone.

The number of autolysosomes, indicating the level of autophagy, is decreased upon co-expression of S6kScer\UAS.cMa and Tak1Scer\UAS.cTa in the developing eye disc, under the control of Scer\GAL4GMR.PF, compared to expression of Tak1Scer\UAS.cTa alone.

Co-expression of P{Sym-UAS-Hsrω} suppresses the eye phenotype of Scer\GAL4GMR.PF, Tak1Scer\UAS.cTa, and also substantially reduces the mortality of differentiated pupae.

Coexpression of HsrωEP93D or HsrωEP3037 enhances the Scer\GAL4GMR.PF, Tak1Scer\UAS.cTa eye phenotype, though pupal lethality is not increased.

The reduced eye phenotype caused by expression of Rab11N124I.Scer\UAS under the control of Scer\GAL4GMR.PF is enhanced by co-expression of Tak1Scer\UAS.cTa.

Coexpression of Tak1Scer\UAS.cTa, with Traf1EP578, under the control of Scer\GAL4GMR.PF, an increased eye roughness and reduced eye size compared to expression of just Traf1EP578.

The addition of BacA\p35GMR.PH to Tak1Scer\UAS.cTa,Scer\GAL4GMR.PF flies suppresses the cell death phenotype in the eye, but has no effect on the abnormal photoreceptor clustering seen in the eye disc. The addition of hep1/Y to Tak1Scer\UAS.cTa,Scer\GAL4GMR.PF flies the reduced eye size phenotype is rescued almost to wild-type. The addition of bsk2/+ to Tak1Scer\UAS.cTa,Scer\GAL4GMR.PF flies the reduced eye size phenotype is rescued almost to wild-type.

Expression of BacA\p35Scer\UAS.cHa does not suppress the neuroblast loss seen in larval brains expressing Tak1Scer\UAS.cTa under the control of Scer\GAL4wor.PA.

Despite suppression of apoptosis through expression of BacA\p35Scer\UAS.cHa, a rough, impaired adult eye phenotype is found in BacA\p35Scer\UAS.cHa/Scer\GAL4GMR.PF; Tak1Scer\UAS.cTa flies, indicating that the Tak1-induced impaired eye phenotype is mainly due to autophagy.
