Expression of nosαTub84B.3'UTR.UAS under the control of Scer\GAL4tj-NP1624 leads to lethality; if the expression is only during adulthood (by the additional control of Gal80[ts] and temperature shift from 18[o]C to 29[o]C), it results in egg chambers with supernumerary germ cells or oocytes.
Animals expressing nosαTub84B.3'UTR.Scer\UAS under the control of Scer\GAL4ppk.PG display severe dendritic arborization defects (loss of terminal branches, decreased total branch length) in class IV dendritic arborizing neurons at third instar larval stage and a significant delay in dendritic pruning of these neurons at the pupal stage.
Over-expression of nosαTub84B.3'UTR.Scer\UAS driven by Scer\GAL4RapGAP1-OK6 results in boutons with irregular rounded shapes and a slight increase in bouton numbers at the third instar larval neuromuscular junction (NMJ), compared to controls; expression driven by Scer\GAL4how-24B or Scer\GAL4C57 produces dramatic changes in NMJ morphology, with abnormal clustering of boutons (not possible to count). Expression driven by Scer\GAL4RapGAP1-OK6 also results in a significant decrease in the number of active zones in type 1s (but not 1b) boutons. Over-expression of nosαTub84B.3'UTR.Scer\UAS driven by Scer\GAL4Mhc.PU results in a significant increase in GluRIIA immunoreactivity.
Expression of nosαTub84B.3'UTR.Scer\UAS under the control of Scer\GAL4221 in class I dendritic arborization neurons of third instar larvae does not dramatically change dendrite morphology compared to controls. Expression of nosαTub84B.3'UTR.Scer\UAS under the control of Scer\GAL4109(2)80 results in a dramatic change in the dendrite morphology of class III dendritic arborization neurons of third instar larvae; the number of high-order dendritic branches is significantly reduced while the morphology of the major branches is not affected. Expression of nosαTub84B.3'UTR.Scer\UAS under the control of Scer\GAL4477 results in a dramatic change in the dendrite morphology of class IV dendritic arborization neurons of third instar larvae; the number of high-order dendritic branches is significantly reduced while the morphology of the major branches is not affected. Expression of nosαTub84B.3'UTR.Scer\UAS under the control of Scer\GAL48-123 results in a reduction in the amount of high-order dendritic branches in class IV dendritic arborization neurons of third instar larvae.
When expression is driven by Scer\GAL4OK107, nosαTub84B.3'UTR.Scer\UAS shows a mushroom body Class III phenotype; axonal growth and guidance defects.
100% of flies expressing nosαTub84B.3'UTR.Scer\UAS under the control of Scer\GAL4GMR.PF have rough eyes. These flies have black spots. 100% of flies expressing nosαTub84B.3'UTR.Scer\UAS under the control of Scer\GAL4hs.2sev have rough eyes. Flies expressing nosαTub84B.3'UTR.Scer\UAS under the control of Scer\GAL4elav.PLu show 82% lethality. Flies expressing nosαTub84B.3'UTR.Scer\UAS under the control of Scer\GAL41407 show 68% lethality. 26% of flies expressing nosαTub84B.3'UTR.Scer\UAS under the control of Scer\GAL4hs.2sev show an "adolescent" phenotype; the wing expansion and body cuticle hardening that normally occurs shortly after eclosion fails to occur. The flies are fertile, but show reduced viability. A number of other Scer\GAL4 lines produce this "adolescent" phenotype in combination with nosαTub84B.3'UTR.Scer\UAS, they are: Scer\GAL4sca-109-68 (74% penetrance), Scer\GAL4HD23A (85% penetrance), Scer\GAL4HD44A (90% penetrance), Scer\GAL4KF42A (38% penetrance) and Scer\GAL4Eh.2.4 (70% penetrance). Flies carrying nosαTub84B.3'UTR.Scer\UAS, Scer\GAL4sca-109-68 and Scer\GAL80αTub84B.PL do not show the adolescent phenotype. Flies carrying nosαTub84B.3'UTR.Scer\UAS, Scer\GAL4sca-109-68 and Scer\GAL80Eh.2.4 do show the adolescent phenotype.
Scer\GAL4HD34A, nanosαTub84B.3'UTR.UAS has lethal phenotype, enhanceable by pum680
Scer\GAL4109(2)80, nanosαTub84B.3'UTR.UAS has abnormal neuroanatomy phenotype, suppressible | partially by pum1/pum1
Scer\GAL4109(2)80, nanosαTub84B.3'UTR.UAS has dendritic arborising neuron & dendritic tree phenotype, suppressible | partially by pum1/pum1
Scer\GAL4sca-109-68, nanosαTub84B.3'UTR.UAS has wing phenotype, non-suppressible by pum680
Scer\GAL4sca-109-68, nanosαTub84B.3'UTR.UAS has adult cuticle phenotype, non-suppressible by pum680
The reduction in high-order dendritic branches seen in class IV dendritic arborization neurons of third instar larvae expressing nosαTub84B.3'UTR.Scer\UAS under the control of Scer\GAL4477 is not as extreme if the larvae are also homozygous for pum1.
The frequency of the adolescent phenotype in animals expressing nosαTub84B.3'UTR.Scer\UAS under the control of Scer\GAL4sca-109-68 is not altered if they are also carrying pum13.
