Description

The three Arg residues immediately upstream of the putative processing site of Actβ (residues 563-565) are mutated to Ala, creating a cleavage mutant. This is driven by UASt sequences.

Ubiquitous expression of two copies of ActβCM.Scer\UAS with Scer\GAL4da.G32 results in smaller animals with wings that are on average 20% smaller than those of wild-type controls.

Expression of ActβCM.Scer\UAS under the control of Scer\GAL4A9 increases the average wing size to 134% that of wild-type, although no obvious pattern defects are observed.

Expression of ActβCM.Scer\UAS specifically in the developing retina under the control of Scer\GAL4GMR.PF results in R7 tiling defects.

When activin-βCM.Scer\UAS is driven by Scer\GAL4αTub84B.PL CNS development is retarded, poor development is seen in the optic lobe. When driven by Scer\GAL4OK107 CNS development is not retarded. Expression of activin-βCM.Scer\UAS when driven by Scer\GAL4hs.PB and heat shocked around the mid-third instar stage blocks optic lobe development.

Co-expression of ActβCM.Scer\UAS and MadScer\UAS.cNa, both under the control of Scer\GAL4A9 dramatically enhances the ectopic wing vein formation seen in MadScer\UAS.cNa mutants alone.

Co-expression of ActβCM.Scer\UAS and SmoxScer\UAS.cGa, both under the control of Scer\GAL4A9 results in mild levels of ectopic wing vein formation.

Expression of ActβCM.Scer\UAS in a dawaact1 background under the control of the neurosecretory driver Scer\GAL4386Y fails to suppress dawaact1 lethality.

Although modest, the tiling phenotype found in ActβCM.Scer\UAS mutants is considerably enhanced when neighboring R7s are removed. Approximately 36% of mutant R7s extend laterally into unoccupied columns.
