Expression of twsScer\UAS.cBa under the control of either Scer\GAL4elav.PU or Scer\GAL4Act5C.PU results in a large number of apoptotic cells in embryos.

Expression of twsScer\UAS.cBa under the control of Scer\GAL4elav.PU reduces the eclosion rate to approximately 40%.

Flies expressing twsScer\UAS.cBa under the control of Scer\GAL4elav.PU at 29[o]C show a severe reduction in lifespan compared to controls.

Newly eclosed adults expressing twsScer\UAS.cBa under the control of Scer\GAL4elav.PU at 29[o]C do not show abnormalities in the brain. However, 7 day old flies show vacuolisation of both cortex and neuropil.

Mitochondrial size is reduced and mitochondrial density is increased in the axons of the larval ventral ganglia in animals expressing twsScer\UAS.cBa under the control of Scer\GAL4en.PU. The mitochondrial reticulum is disintegrated into multiple punctiform organelles.

Mitochondria of photoreceptor neurons are considerably smaller than controls in flies expressing twsScer\UAS.cBa under the control of Scer\GAL4elav.PU. Scission of mitochondria is seen. Internal cristae of disintegrated mitochondria are less condensed than normal and some are circled by autophagosome-like organelles.

Mitochondrial membrane potential is significantly reduced in the heads of flies expressing twsScer\UAS.cBa under the control of Scer\GAL4elav.PU.

Flies expressing twsScer\UAS.cBa under the control of Scer\GAL4elav.PU at 29[o]C and treated with 20mM paraquat show reduced climbing ability compared to control flies.

twsScer\UAS.cBa; Scer\GAL4hs.PB; tws60/tws60 animals have scattered ectopic bristles on their wing blades.

Genetic Interactions

Co-expression of Sod2Scer\UAS.cMa extends the lifespan of flies expressing twsScer\UAS.cBa under the control of Scer\GAL4elav.PU at 29[o]C.

Xenogenetic Interactions
Statement
Reference

Expression of twsScer\UAS.cBa mitigates the Hsap\MAPTScer\UAS.cWa-induced bristle loss phenotype (with both under the control of Scer\GAL4Eq1). Bristle and sensory cell numbers increase concurrently when twsScer\UAS.cBa is co-expressed with Hsap\MAPTScer\UAS.cWa.

Expression of twsScer\UAS.cBa in larval da neurons (under the control of Scer\GAL4ppk.PG) suppresses the aggregate formation and dysmorphology of neurites found upon expression of Hsap\MAPTScer\UAS.cWa.

Comments

twsScer\UAS.cBa; Scer\GAL4hs.PB rescues survival of tws60 to the pharate adult stage, with no obvious morphological defects apart from scattered ectopic bristles on the wing blades. The larval/pupal lethality of twsj11C8 homozygotes is not rescued by twsScer\UAS.cBa; Scer\GAL4hs.PB, but is partially rescued by twsScer\UAS.cBa; Scer\GAL4en-e16E.
