Overexpression of gwlL.Scer\UAS in eggs/embryos under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16 from twsj11C8/+ mothers is almost completely lethal. Most eggs/embryos are blocked in metaphase of meiosis I, even after 4 to 6 hours post-laying. Only a few eggs manage to complete meiosis and attempt to initiate embryonic development. However, these embryos usually abort in the first mitosis, displaying several aberrant structures with condensed chromatin at the center of small spindles. The few mitotic nuclei observed show a very high incidence of detached centrosomes.
Female germline expression of gwlL.Scer\UAS (under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16) in polo11 heterozygous females allows partial egg hatch (around 4%), and these embryos also show loss of centrosomes from spindles in early syncytial divisions.
Expression of gwlL.Scer\UAS under the control of Scer\GAL4Act5C rescues the viability and somatic integrity of gwlSr3/gwlSr6, gwlSr3/gwlSr3, gwlSr3/Df(3L)gwl3b and gwlSr3/Df(3L)gwl3b flies to similar degrees.
The fertility of gwlSr18/Df(3L)gwl3b females is rescued completely by expression of gwlL.Scer\UAS in the maternal germline with Scer\GAL4mat.αTub67C.T:Hsim\VP16.
