Description

UASt regulatory sequences drive expression of Hsap\MAPT, carrying mutations in fourteen disease-associated Ser-Thr-Pro sites (Thr 111, Thr 153, Ser 175, Thr 181, Ser 199, Ser 202, Thr 205, Thr 212, Thr 217, Thr 231, Ser 235, Ser 396, Ser 404, and Ser 422) to glutamate.

4 week old flies expressing Hsap\MAPTE14.Scer\UAS under the control of Scer\GAL4ple.PF show a significant loss of PPL1 dopaminergic neurons in the brain. No significant loss of these neurons is seen in younger flies.

The mitochondria in the neurons of adult brains expressing Hsap\MAPTE14.Scer\UAS under the control of Scer\GAL4elav.PU are markedly elongated. Mitochondria length is on average greater than twice that of controls.

Overexpression of Hsap\MAPTE14.UAS under the control of Scer\GAL4RapGAP1-OK6 in larval segmental nerves effects synaptic vesicle kinetics (there is a significant increase in instant velocity); there is no significant reduction in MT density; no effect on the number of mobile vesicles within axons. Emerging flies have normal wing expansion.

Compared to controls, expression of Hsap\MAPTE14.Scer\UAS under the control of Scer\GAL4elav.PU results in severe mushroom body (MB) abnormalities. However, although the number of MB neurons is severely reduced, they are not completely eliminated.

Expression of Hsap\MAPTE14.Scer\UAS in the developing eye under the control of Scer\GAL4GMR.PF generates a severe rough eye phenotype. The orderly ommatidial architecture is disorganized and photoreceptors are fused with missing mechanosensory bristles.

Expression of Hsap\MAPTE14.Scer\UAS in the developing external sensory organs under the control of Scer\GAL4Eq1 generates a moderate degree of bristle loss.

Expression of Hsap\MAPTE14.Scer\UAS under the control of Scer\GAL4GMR.PF results in a severe rough eye phenotype.

Expression of Hsap\MAPTE14.Scer\UAS under the control of Scer\GAL4elav.PU results in a rough eye phenotype.

Conditional expression of Hsap\MAPTE14.Scer\UAS only during the adult stages under the control of Scer\GAL4elav.PU and Scer\GAL80ts.αTub84B substantially and significantly elongates the lifespan of flies compared with wild-type.

Expression of Hsap\MAPTE14.Scer\UAS under the control of Scer\GAL4elav.PU results in neurodegeneration of both the cortex and neuropil in 5-day old flies, with these regions exhibiting an increase in neuron apoptosis, as compared to controls.

Expression of Hsap\MAPTE14.Scer\UAS in the retina under the control of Scer\GAL4GMR.PF results in a rough-eye phenotype.

Expression of Hsap\MAPTE14.Scer\UAS under the control of Scer\GAL4GMR.PF results in a rough-eye phenotype.

Expression of Hsap\MAPTE14.Scer\UAS in the brain under the control of Scer\GAL4elav.PLu results in neurodegeneration, and increased cell cycle activation is also observed.

Either Trxr-1Δ1 heterozygotes or Sod2n283 heterozygotes expressing Hsap\MAPTE14.Scer\UAS under the control of Scer\GAL4elav.PU are short lived as compared to controls.

Expression of Act5CScer\UAS.T:Avic\GFP in animals expressing Hsap\MAPTE14.Scer\UAS in the retina under the control of Scer\GAL4GMR.PF enhances the rough-eye phenotype.

The rough-eye phenotype observed in animals expressing Hsap\MAPTE14.Scer\UAS in the retina under the control of Scer\GAL4GMR.PF is enhanced in a Act5CG0010/+ background.

Expression of Hsap\APPAβ42.Scer\UAS in flies expressing Hsap\MAPTE14.Scer\UAS under the control of Scer\GAL4GMR.PF does not enhance the rough-eye phenotype.

The rough-eye phenotype observed in animals expressing Hsap\MAPTE14.Scer\UAS under the control of Scer\GAL4GMR.PF is suppressed by co-expression of dapScer\UAS.cdNa and RbfScer\UAS.cDa.

The rough-eye phenotype observed in animals expressing Hsap\MAPTE14.Scer\UAS under the control of Scer\GAL4GMR.PF is enhanced by co-expression of CycDScer\UAS.cMa.

The rough-eye phenotype observed in animals expressing Hsap\MAPTE14.Scer\UAS under the control of Scer\GAL4GMR.PF is enhanced by co-expression of CycEScer\UAS.cUa.
