Description

In the absence of a GAL4 driver, the P{UAS-DTHg} construct shows complete rescue of flies that lack ple function in the central nervous system. This is most likely due to expression driven by intragenic transcriptional regulatory elements (the ple sequence in this construct is a genomic fragment including introns).

UASt regulatory sequences drive expression of the whole transcribed region of ple.

Expression of pleScer\UAS.cFa under the control of Scer\GAL4ple.PF results in increased resistance of flies to paraquat intoxication. Increased survival rates are seen at 24 hours after ingestion and 2 hours after direct application to the ventral nerve cord (VNC) of decapitated flies.

Expression of pleScer\UAS.cFa in serotonergic neurons under the control of Scer\GAL4Trh.PB results in increased resistance of flies to paraquat intoxication. Increased survival rates are seen at 24 hours after ingestion and 2 hours after direct application to the ventral nerve cord (VNC) of decapitated flies.

Flies expressing pleScer\UAS.cFa in a ple2 background (under the control of both Scer\GAL4ple.PF and Scer\GAL4Ddc.PL) display slightly less accurate visual orientation than wild-type. Similarly, using the detour paradigm, both pleScer\UAS.cFa; ple2 and pleFS.Scer\UAS; ple2 flies demonstrate a roughly wild-type spatial orientation memory.

ple2 flies expressing either pleScer\UAS.cFa under the control of both Scer\GAL4ple.PF and Scer\GAL4Ddc.PL exhibit normal adult life expectancy.

Flies expressing pleScer\UAS.cFa in a ple2 background (under the control of both Scer\GAL4ple.PF and Scer\GAL4Ddc.PL) demonstrate associative memory similar to wild-type, indicating normal olfactory learning.

Flies expressing pleScer\UAS.cFa in a ple2 background (under the control of both Scer\GAL4ple.PF and Scer\GAL4Ddc.PL) demonstrate partial rescue of ple2 phototactic behavior.
