UAS regulatory sequences drive expression of a short inverted repeat.
The expression of Lis-1HMS01457 under the control of Scer\GAL4shot-OK307 or Scer\GAL4GMR91H05, but not under the split-GAL4 Hsap\RELAAD.R17A04 + Scer\GAL4DBD.R68A06 combo, results in shorter and thinner giant fiber terminals and disrupted electrophysiological connection to TTMn (as shown by a severely increased latency and a severely decreased ability to follow stimuli), as compared to controls. Expression under the control either of Scer\GAL4GMR68A06 or of the split-Gal4 Hsap\RELAAD.R17A04 + Scer\GAL4DBD.R68A06 combo leads to a severe impairment of the axonal transport of Nrg-positive vesicles in giant fibers, as compared to controls.
Lis-1HMS01457 is rescued by Lis-1UAS.Emerald/Scer\GAL4shot-OK307
The giant fiber anatomical and electrophysiological defects induced by the expression of Lis-1HMS01457 under the control of Scer\GAL4shot-OK307 are rescued by the co-expression of Lis-1Scer\UAS.T:Avic\GFP-EMD.
