The pUASg-HA.attB plasmid is designed for generating donor plasmids that can be used to insert sequence containing an open reading frame (ORF) of interest, expressed under the control of UASt regulatory sequences and tagged at the C-terminal end with three copies of Tag:HA, into an attP-containing docking site in the genome, via phiC31:int-mediated integration. An ORF of interest can be inserted into the plasmid via a Gateway cassette.
pUASg-HA.attB contains the w+mC marker; Scer\UAS sequences followed by a Gateway cassette integration site (allowing insertion of the ORF of choice) followed by a C-terminal 3xHA (Ivir\T:Ivir\HA1) epitope tag; and a Sliv\attB site, allowing use as a donor for the phiC31-based integration system.
