Detailed Description

Expression of amosScer\UAS.cGa under the control of Scer\GAL4dpp.blk1 results in a significant increase in the size of the wing disc. Ectopic sensory organ precursor cells are formed in the disc. These animals occasionally develop to pharate adults at 18oC and in this case they have a large number of ectopic macrochaetae. Expression of amosScer\UAS.cGa under the control of Scer\GAL4hs.PB using a 6 minute heat shock at 38oC results in the induction of mechanosensory organs and campaniform sensilla in the wing.

Animals expressing amosScer\UAS.cGa under the control of Scer\GAL4C-765 can develop to pharate adults at 17oC. Their bodies are covered with many sensory organs of different types. These are mostly macrochaetae on the head and on the notum. Large numbers of sensilla campaniformia appear in the proximal wing and the rest of the wing contains a mixture of bristles of diverse types and sensilla campaniformia.

When amosScer\UAS.cGa is driven by Scer\GAL4sca-109-68 there is a dramatic reduction in bristle formation.

Flies expressing amosScer\UAS.cGa under the control of Scer\GAL4GMR.12 have no eye defect. Flies expressing amosScer\UAS.cGa under the control of Scer\GAL4hs.PB (expressed using a 30 minute 37oC heat shock at the beginning of the third larval instar) results in a rough eye phenotype. Third instar eye discs have irregularly spaced R8 precursors and occasional R8 clusters.

Expression of amosScer\UAS.cGa under the control of Scer\GAL4OK384 results in a dramatic increase in olfactory sensilla, with an increase of 24-44% for all sensillum types. A proportion of the sensilla appear to be of intermediate basiconica/trichodea morphology (they are shaped like sensilla basiconica but are more pigmented like sensilla trichodea). The antenna is bloated. Expression of amosScer\UAS.cGa under the control of Scer\GAL4sca-109-68 results in a mixture of ectopic sense organs. Ectopic chordotonal organs are formed in the scutellum and third wing vein. Some extra external sense organs are formed, mostly along the third wing vein. In addition, unusual organs that appear morphologically similar to olfactory sensilla (particularly sensilla trichodea) are induced in the thorax, head and along the third wing vein. At high expression levels of amos, some of the sensory bristles of the thorax are transformed to olfactory-like morphology; while wild-type thoracic bristles are innervated by a single sensory neuron, a small proportion of these transformed sensilla have two or more sensory neurons, a characteristic of olfactory sensilla. Expression of amosScer\UAS.cGa under the control of Scer\GAL4hs.PB results in a large number of ectopic olfactory-like sensilla on the antenna.

The reduction in number of sensilla basiconica seen in lz3 flies is partially rescued by expression of amosScer\UAS.cGa under the control of Scer\GAL4hs.PB.

Comments

Expression of amosScer\UAS.cGa under the control of Scer\GAL4amos.PZ substantially rescues the mutant phenotype seen in the sensilla of the third antennal segment in amos1/Df(2L)M36F-S6 flies; ectopic bristles are almost completely suppressed in the rescued flies, and sensilla trichodea are present in numbers close to wild type. Substantial numbers of sensilla basiconica are also present in the rescued flies, but the number is less than half that expected for wild type.
