Expression of NcScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4Crz.504 results in apoptosis in all vCrz neurons, but not in proto-cerebral corazonergic neurons.

Approximately 95% of larvae expressing Scer\GAL4Adh.PF-driven NcScer\UAS.T:Avic\GFP-EGFP in the fat body die during different larval stages. The remaining animals die at the pupal stage. A significant portion of fat body cells in these mutants display apoptosis at the early wandering third instar larval stage.

Expression of NcScer\UAS.T:Avic\GFP-EGFP, under the control of Scer\GAL4elav-C155, shortens lifespan in flies.

NcScer\UAS.T:Avic\GFP-EGFP; Scer\GAL4hs.PB rescues pupariation of most Nc30, Nc11 or Nc51 homozygous larvae. It also rescues cell death in the larval midgut in response to feeding mid-third instar larvae with ecdysone and the normal pattern of cell death of abdominal neuroblasts in the late third instar.

When NcScer\UAS.T:Avic\GFP-EGFP is driven by Scer\GAL4GMR.PF, animals have small mottled eyes.

Expression of NcScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4GMR.PF results in massive induction of cell death in the posterior part of the eye imaginal disc. Expression of NcScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4hs.PB during embryogenesis or larval development using heat shock results in ectopic cell death. Many animals expressing NcScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4GMR.PF die as pupae. The few adults that survive have severely ablated eyes. Insertions of P{UAS-Nc.EGFP} that result in less lethality when expressed under the control of Scer\GAL4GMR.PF give rise to adult flies with slightly rough and mottled eyes at 25oC. The phenotype is more severe at 29oC.

Wing discs coexpressing BacA\p35Scer\UAS.cHa and NcScer\UAS.T:Avic\GFP-EGFP under the control of either Scer\GAL4en-e16E or Scer\GAL4ptc-559.1 show increased cell proliferation in the domain of Scer\GAL4 expression.

Xenogenetic Interactions
Statement
Reference

Co-expression of BacA\p35Scer\UAS.cHa with NcScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4Crz.504 does not prevent the Nc-induced apoptosis in vCrz neurons.

Expression of NcScer\UAS.T:Avic\GFP-EGFP and BacA\p35Scer\UAS.cHa in the wing disc, driven by Scer\GAL4en-e16E, induces a strong overproliferation phenotype.

Comments

NcScer\UAS.T:Avic\GFP-EGFP; Scer\GAL4hs.PB rescues pupariation of most Nc30, Nc11 or Nc51 homozygous larvae. It also rescues cell death in the larval midgut in response to feeding mid-thrd instar larvae with ecdysone and the normal pattern of cell death of abdominal neuroblasts in the late third instar.
