Elimination of molecular clock function (using cycΔ.UAS.Tag:MYC) in Scer\GAL4Clk.2341 cells in adults significantly reduces the strength of the circadian rhythm of sleep-wake cycles, compared to controls; this is also the case when expression is restricted to a smaller subset of cells using Scer\GAL80Pdf.PS.

Inhibition (using Hsap\KCNJ2UAS.EGFP) of Scer\GAL4Clk.2341 cells significantly reduces the strength of the circadian rhythm of sleep-wake cycles, compared to controls; this is also the case when inhibition is restricted to a smaller subset of cells using Scer\GAL80Pdf.PS and when inhibition of this subset is restricted to the adult stage using a Gal80[ts] transgene.

Optogenetic activation (using Cnoc\ChR1::Csub\ChRCsChrimson.20xUAS.FRT.Venus with Scer\FLP1L.8xlexAop and 627 nm (red) light) of cells expressing Scer\GAL4Clk.2341 with Ecol\lexAGMR19C05 or Ecol\lexAGMR43D05 in adults significantly reduces sleep, compared to controls; activation of cells expressing Scer\GAL4Clk.2341 with Ecol\lexAGMR73B06, Ecol\lexAGMR64A11, Ecol\lexAGMR81E08, Ecol\lexAGMR55C09, Ecol\lexAGMR19A11, Ecol\lexAGMR70E02 or Ecol\lexAGMR70G01 in adults does not significantly affect sleep, compared to controls; activation of cells expressing Scer\GAL4Clk.2341 with Ecol\lexAGMR66B05, Ecol\lexAGMR77H08, Ecol\lexAGMR12G04, Ecol\lexAGMR67F03, Ecol\lexAGMR65D05 or Ecol\lexAGMR18H11 in adults significantly increases sleep, compared to controls; activation of a subset of Scer\GAL4Clk.2341 and Ecol\lexAGMR16C05-expressing cells (restricted using Scer\GAL80cry.PS) in adults does not significantly affect sleep, compared to controls; activation of a subset of Scer\GAL4Clk.2341 and Ecol\lexAGMR18H11-expressing cells (restricted using Scer\GAL80cry.PS) in adults significantly reduces sleep, compared to controls.
