20xUAS drives expression of CsChrimson fused to Venus. Golgi and ER export motifs have also been added to the coding sequence. FlyBase curator comment: the original published version of FBrf0224260 erroneously stated that this construct contained Chrimson rather than CsChrimson; this has since been corrected in the online published version (for details see also the corrigendum FBrf0226484 and the addendum FBrf0227011).

In larvae expressing Zzzz\ChR1IVS.20xScer\UAS.T:Avic\GFP-YFP.Venus in motor neurons (via Scer\GAL4VGlut-OK371), both 470-nm and 617-nm light triggers EJPs, even at short (1- to 2-ms) light-pulse durations and low intensities. In response to light-pulse durations over 2 ms, Zzzz\ChR1IVS.20xScer\UAS.T:Avic\GFP-YFP.Venus expression triggers long-lasting barrages of EJPs. Long-wavelength red light (720 nm) also triggers EJPs, though high-intensity light and long light-pulse durations (40–160 ms) are required for robust activation of the NMJ.

In adults expressing Zzzz\ChR1IVS.20xScer\UAS.T:Avic\GFP-YFP.Venus in sweet taste receptor cells (via Scer\GAL4Gr64f.9.7), proboscis extension reflexes are robustly elicited at very low 470 nm and 617 nm light intensities. Flies also respond to 720-nm light at an intensity substantially lower than that required in the larvae.

Adults expressing Zzzz\ChR1IVS.20xScer\UAS.T:Avic\GFP-YFP.Venus in PNv-1 antennal lobe neurons that innervate the V glomerulus (via Scer\GAL4VT31497) which induces an avoidance response to CO[[2]], reliably avoid quadrants in a circular arena lit by weak red light (617 nm).
