Description

Imprecise excision of the progenitor insertion has resulted in a deletion which affects both Gyc76C and HLH106 (neighbouring genes). The deletion extends 697bp into the HLH106 open reading frame, up to amino acid residue 233 in exon 3 (the next in-frame start codon is at amino acid residue 265). The deletion also removes the predicted first exon of Gyc76C.

Comment:

Deletion extends from site of P{SUPor-P}Gyc76CKG03723 insertion within Gyc76C to amino acid 233 of SREBP. The deletion removes 697 nucleotides of the SREBP ORF and the first noncoding exon of Gyc76C.

Detailed Description

HLH106189 mutants survive extremely poorly on unsupplemented medium, and their survival is much improved on medium supplemented with fatty acids.

In HLH106189 mutant flies, lethality is reduced by supplementing food with fatty acids.

Homozygous larvae show a profound defects in growth during the second larval instar.

HLH10652/HLH106189 first instar larvae contain significantly less total fatty acid than wild-type controls, but the relative abundance of various fatty acid species does not differ significantly from that of controls.

The survival of homozygotes to adulthood is markedly improved in the presence of soy lipids (lecithin) in the food medium, and increases with increasing concentration of soy lipid (up to 9%). The rescued adults are indistinguishable in mass from their heterozygous siblings and are morphologically normal.

A SREBP189 heterozygous background is sufficient to ameliorate the lipid droplet accumulation displayed in sicilyE mutants.

A SREBP189 heterozygous background is sufficient to ameliorate the lipid droplet accumulation displayed in Aats-metFB mutants.

A SREBP189 heterozygous background is sufficient to ameliorate the lipid droplet accumulation displayed in MarfB mutants.

On unsupplemented medium, S2PKG08356/S2P1 mutants in the presence of two copies of HLH106D386A and carrying HLH106189/+, survive at less than 25% of the expected rate. When these animals are homozygous for HLH106189, survival at only 10% of the expected rate is observed. Survival is restored by supplementing the culture medium with free fatty acids.
