Detailed Description

The expression of Sec71HMS00357 under the combined control of Scer\GAL4ppk.PU and Dicer-2 transgene (for efficient RNAi), leads to dorsal multidendritic ddaC neurons exhibiting a significantly simplified dendritic tree at the white pupa stage and a fully penetrant delay in pruning at 16h-18h after puparium formation, as compared to controls; at the wandering third instar larval stage, these neurons exhibiting defects in the formation of secretory vesicles, as shown by the virtual absence of punctate structures for the exocyst marker Sec15, as compared to controls.

Expression via Scer\GAL4fkh.PU results in larval salivary glands that have significantly smaller secretory granules, compared to controls.

Both the simplified dendritic arborization at the white pupa stage and the pruning defects at 16h-18h after puparium formation observed in dorsal multidendritic ddaC neurons expressing Sec71HMS00357 under the combined control of Scer\GAL4ppk.PU and Dicer-2 transgene are suppressed by the co-expression of Arf79FScer\UAS.RNAi-resistant; the pruning defects at 16h after puparium formation are also partially suppressed by the co-expression of either NrgGL00656 or NrgHMS01638, but not Arf79FQ71L.Scer\UAS.cWa or Arf51FScer\UAS.cCa.

Comments

Both the dendritic arborization defects at the white pupa stage and the dendrite pruning defects at 16h after puparium formation observed in dorsal multidendritic ddaC neurons upon the expression of Sec71HMS00357 under the combined control of Scer\GAL4ppk.PU and Dicer-2 transgene are fully rescued by the co-expression of Sec71Scer\UAS.RNAi-resistant, Sec71ΔHDS2-4.Scer\UAS or Sec71ΔHDS3-4.Scer\UAS; the co-expression of Sec71ΔDCB.Scer\UAS, however, only partially rescues the dendritic arborization defects at the white pupa stage, but not the pruning defects at 16h after puparium formation.
