Females with AlkGD42 (along with Dcr2) driven by Scer\GAL4elav.PU, Scer\GAL4ey-OK107, Scer\GAL4H24, Scer\GAL4386Y, Scer\GAL4007Y, Scer\GAL4c453, Scer\GAL4MJ63, Scer\GAL41471, Scer\GAL430Y, Scer\GAL4121Y, Scer\GAL4c320, Scer\GAL4NP1131, Scer\GAL4238Y, Scer\GAL4ChAT.7.4 or Scer\GAL4nSyb.PS sleep significantly more than controls; when expression is driven by Scer\GAL4c309, females sleep significantly less than controls. Females with AlkGD42 (along with Dcr2) driven by Scer\GAL4386Y, Scer\GAL41471 and Scer\GAL4c320 mainly show increases in daytime sleep, whereas those with expression driven by Scer\GAL4007Y mainly show increases in nighttime sleep. Females with AlkGD42 (along with Dcr2) driven by Scer\GAL4GMR71G10, Scer\GAL4GMR76D11, Scer\GAL417d, Scer\GAL4D52H or Scer\GAL4c305a do not show significant changes in sleep amount compared to wild type.

Using Scer\GAL80Mef2.mb247 to exclude AlkGD42 expression from mushroom bodies eliminates the sleep increase induced by Scer\GAL4386Y or Scer\GAL430Y, and in the case of Scer\GAL430Y even significantly decreasing total sleep compared to wild type; the decrease in sleep seen with Scer\GAL4c309 is further significantly decreased.

Pan-neuronal expression of AlkGD42 under the control of Scer\GAL4elav-C155, and restricted to the adult stages using Scer\GAL80ts.αTub84B, causes a reproducible increase in associative olfactory learning performance compared to controls.

Expression of AlkGD42 under the control of Scer\GAL4Ras64B.PW, and restricted to the adult stages using Scer\GAL80ts.αTub84B, causes a reproducible increase in associative olfactory learning performance compared to controls. This increase in performance is seen when flies are trained with 3 conditional stimulus (CS)/ unconditioned stimulus (US) pairings, and a further enhancement in learning ability is seen with 6 pairings. Training with 12 CS/US pairings does not enhance the phenotype any further. The wings of the resulting adult flies are also larger than in wild type, due to increased cell size rather than cell number.

Expression of AlkGD42 under the control of Scer\GAL4Alk.38, and restricted to the adult stages using Scer\GAL80ts.αTub84B, causes a reproducible increase in associative olfactory learning performance compared to controls. This increase in performance is seen when flies are trained with 3 conditional stimulus (CS)/ unconditioned stimulus (US) pairings.

Expression of AlkGD42 under the control of either Scer\GAL4Mef2.247 or Scer\GAL4c747, and restricted to the adult stages using Scer\GAL80ts.αTub84B, does not affect associative olfactory learning.

Pan-neuronal expression of AlkGD42 under the control of Scer\GAL4elav-C155 throughout development yields pupae that are 10-15% larger than controls. The resulting adults are viable and normally patterned.

Expression of AlkGD42 under the control of Scer\GAL4Ras64B.PW throughout development yields pupae that are 10-15% larger than controls.

Expression of AlkGD42 under the control of Scer\GAL4Alk.38 throughout development yields pupae that are 10-15% larger than controls.

Expression of AlkGD42 under the control of Scer\GAL4386Y throughout development yields pupae that are 10-15% larger than controls.

Expression of AlkGD42 under the control of Scer\GAL4Ilp2.PR throughout development yields pupae that are a similar size to controls.

Adults expressing AlkGD42 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Nf1P1/Nf1P2 suppresses the long sleep phenotype seen in flies with AlkGD42 driven by Scer\GAL4nSyb.PS (at 25[o]C in female flies).

Expression of AlkGD42 under the control of Scer\GAL4elav-C155 rescues the small pupae phenotype seen in Nf1E2 homozygotes. The pupae seen are close to the size of controls. Rescue is also seen when AlkEC.Scer\UAS.T:Hsap\MYC is expressed under the control of Scer\GAL4Ras64B.PW but the rescue is not as efficient. The increase in size is due to increased cell size rather than increased cell number. Expression of AlkGD42 under the control of Scer\GAL4386Y also partially rescues the phenotype in Nf1E2.

Expression of AlkGD42 under the control of Scer\GAL4ey-OK107 does not suppress the small pupae phenotype seen in Nf1E2 mutants.

Expression of AlkGD42 under the control of Scer\GAL4Ccap.PP does not suppress the small pupae phenotype seen in Nf1E2 mutants.

Expression of AlkGD42 under the control of either the Scer\GAL4elav-C155, Scer\GAL4Ras64B.PW or Scer\GAL4Alk.38 driver, and restricted to the adult stages using Scer\GAL80ts.αTub84B, rescues the olfactory learning defects seen in Nf1E2/Nf1E1 mutants.

Expression of AlkGD42 under the control of Scer\GAL4c747, and restricted to the adult stages using Scer\GAL80ts.αTub84B, fails to rescue the olfactory learning defects seen in Nf1E2/Nf1E1 mutants.
