No dopaminergic neuron loss is observed when Scer\NDI1Scer\UAS.cSa is expressed under the control of Scer\GAL4ple.PF.

Flies with expression of Scer\NDI1Scer\UAS.cSa driven by Scer\GAL4da.G32 are viable and fertile, with no morphological, behavioral or reproductive abnormalities. Scer\GAL4da.G32>Scer\NDI1Scer\UAS.cSa flies show significant resistance to rotenone or paraquat but not antimycin, compared to controls; the range of drug concentration where resistance is seen is narrow and males appear more resistant than females. Scer\GAL4da.G32>Scer\NDI1Scer\UAS.cSa flies have a significantly longer lifespan than controls; this is more pronounced in males than females. Scer\GAL4da.G32>Scer\NDI1Scer\UAS.cSa flies have significantly longer lifespans than controls when raised on rich medium or under dietary restriction conditions. Isolated mitochondria from aged Scer\GAL4da.G32>Scer\NDI1Scer\UAS.cSa flies (in contrast to young flies) show increases in substrate oxidation and decreases in mitochondrial ROS production, compared to controls.

Co-expression of Zzzz\AOXScer\UAS.cFa and Scer\NDI1Scer\UAS.cSa under the control of Scer\GAL4da.G32 in tko25t/tko25t mutants results in complete lethality, and in tko25t/+ mutants results in partial lethality.

Co-expression of Scer\NDI1Scer\UAS.cSa does not rescue lethality in flies in flies with expression of COX4KK107510, COX5BGD6659 or COX7AKK110870 driven by Scer\GAL4da.PU. Co-expression of Scer\NDI1Scer\UAS.cSa leads to a shortened lifespan in females and further enhances the shortened lifespan in males with expression of COX5BGD6659 driven by Scer\GAL4elav-C155. Co-expression of Scer\NDI1Scer\UAS.cSa enhances the shortened lifespan in males with expression of COX4KK107510 driven by Scer\GAL4elav.PLu. Expression of Scer\NDI1Scer\UAS.cSa driven by Scer\GAL4da.PU does not significantly suppress bang sensitivity in levy1 flies. Flies with expression of ND-39GD5150 or ND-19GD16787, along with Scer\NDI1Scer\UAS.cSa, driven by Scer\GAL4da.PU show significantly increased recovery time (bang sensitivity) compared to controls.

Co-expression of Zzzz\AOXScer\UAS.cFa leads to lethality (less than 2% survivors) in flies with expression of Scer\NDI1Scer\UAS.cSa driven by Scer\GAL4da.G32.

Co-expression of Scer\NDI1Scer\UAS.cSa suppresses lethality in flies with expression of ND-19GD16787 or ND-39GD5150 driven by Scer\GAL4da.G32.
